The phytochrome B encoded by the HLG locus of Nicotiana plumbaginifolia is required for detection of photoperiod: hlg mutants show altered regulation of flowering and circadian movement

چکیده
The sensitivity of the Nicotiana plumbaginifolia wild-type and phytochrome B (hlg) mutant to photoperiod was investigated. Leaf number at bolting and rosette-leaf movement were measured under conditions of different light quality and photoperiod. Bolting in wild-type plants was slightly delayed under short days (SD), compared to plants grown under long days (LD). The hlg mutant bolted later than the wild-type under LD, but not under SD. An endogenous rhythm of rosette-leaf movement in the wildtype was present under SD, but suppressed under LD. The hlg mutant behaved indistinguishably from the wild-type under SD conditions, but the movements were not suppressed under LD in the mutants. The endogenous rhythm appeared to be entrained normally in the mutants under all the conditions investigated. It was concluded that the B-type phytochrome absent in the hlg mutant is required for the measurement of photoperiod duration in N. plumbaginifolia, but is not required for entrainment of the circadian clock.

Nicotiana plumbaginifolia hlg mutants have a mutation in a PHYB-type phytochrome gene: they have elongated hypocotyls in red light, but are not elongated as adult plants.
Two new allelic mutants of Nicotiana plumbaginifolia have been isolated which display a hypocotyl which is long (hlg) when seedlings are grown in continuous white light (W). This can be accounted for by the decreased response to red light (R) of the hypocotyl elongation rate in these mutants. Responses to other wavelengths are unaffected in the mutants. When grown in white light, mature hlg mut...
متن کاملEARLY FLOWERING 4 functions in phytochrome B-regulated seedling de-etiolation.
To define the functions of genes previously identified by expression profiling as being rapidly light induced under phytochrome (phy) control, we are investigating the seedling de-etiolation phenotypes of mutants carrying T-DNA insertional disruptions at these loci. Mutants at one such locus displayed reduced responsiveness to continuous red, but not continuous far-red light, suggesting a role ...
متن کاملPhytochrome C plays a major role in the acceleration of wheat flowering under long-day photoperiod.
Phytochromes are dimeric proteins that function as red and far-red light sensors influencing nearly every phase of the plant life cycle. Of the three major phytochrome families found in flowering plants, phytochrome C (PHYC) is the least understood. In Arabidopsis and rice, PHYC is unstable and functionally inactive unless it heterodimerizes with another phytochrome. However, when expressed in ...

To investigate the impact of overexpression of AtNRT2.1 transgene from Arabidopsis on nitrate uptake rate and to understand the regulation of endogenous HATS by nitrate and glutamine amino acid (Gln) in tobacco plants, wild-type and transgenic (F line) plants grown on soil for 4 weeks were transferred to hydroponic culture in a controlled-environment with a 16/8h L:D photoperiod at 24? C/20...
متن کاملPea LATE BLOOMER1 is a GIGANTEA ortholog with roles in photoperiodic flowering, deetiolation, and transcriptional regulation of circadian clock gene homologs.
Genes controlling the transition to flowering have been studied in several species, including Arabidopsis (Arabidopsis thaliana) and rice (Oryza sativa), but have not yet received much attention in legumes. Here, we describe a new allelic series of late-flowering, photoperiod-insensitive mutants in the pea (Pisum sativum) LATE BLOOMER1 (LATE1) gene and show that LATE1 is an ortholog of Arabidop...
